Endometriosis Overview
Endometriosis is defined as the presence of endometrial glands and stroma like lesions outside of the uterus. The lesions can be peritoneal lesions, superficial implants or cysts, or deep infiltrating disease. While there is no definitive etiology of endometriosis, there are several hypotheses regarding how endometriotic lesions develop. One possible mechanism is retrograde, a feature of the menstrual cycle in women and non-human primates, which is an outflow of the endometrial lining through the patent fallopian tubes into the pelvic space. This retrograde flow, along with potential hematogenous or lymphatic circulation, may result in the seeding of endometrial tissue in ectopic sites.

Endometriosis Emerging Drugs
• HMI-115: Hope Medicine (Nanjing) Co., Ltd
HMI-115 is a first-in-class monoclonal antibody drug targeting the prolactin receptor (PRLR), that Hope Medicine owns the global rights. The product candidate is being developed for multiple indications globally. Potential indications for the antibody include Endometriosis (EM), Androgenic Alopecia (AGA), and other chronic diseases. This antibody has shown excellent characteristics in animal models including non-human primate (NHP) models and human safety. Currently the drug is in Phase III stage of its development for the treatment of Endometriosis.

• Merigolix: TiumBio
Merigolix (TU2670) is an oral GnRH antagonist that can bind to pituitary receptors to suppress estradiol hormone. The existing treatment GnRH agonists, lowers patient comfort levels as it's in an injection form and inhibits sex hormones to the level of menopause, resulting in serious side effects such as bone loss. TU2670 is an oral GnRH antagonist that only suppresses hormone levels to the level of efficacy, improving both side effects and ease of administration by oral. In addition, being a sex hormone control mechanism, it allows for indication expansion to precocious puberty, test-tube baby, infertility treatment, etc. The drug is currently being evaluated under Phase II clinical trial for the treatment of endometriosis.

• Nibrozetone (RRx-001): EpicentRx
Nibrozetone, also known as RRx-001, has a unique mechanism of action that involves both anti-inflammatory and antioxidant properties. It has a dual mechanism of action, fragmenting to release metabolites such as nitric oxide (NO) and other nitrogen-centered free radicals in hypoxic environments, targeting diseases like cancer. In non-hypoxic environments, RRx-001 has anti-toxicity activity through direct inhibition of the NLRP3 inflammasome and upregulation of the master antioxidant transcription factor, Nrf2. Additionally, it inhibits the CD47 antigen, involved in immune cell function and tumor progression. Currently, the drug is in the Phase I stage of its development for the treatment of endometriosis.
