What is Thyroid Cancer?
Thyroid cancer is a malignant condition that primarily arises from the follicular cells of the thyroid gland, which are responsible for producing thyroid hormones. The major types of Thyroid Cancer include papillary, follicular, medullary, and anaplastic thyroid cancers, with papillary thyroid cancer being the most common and generally the least aggressive.
Symptoms may include a neck lump, hoarseness, difficulty swallowing, and swollen lymph nodes, although many early cases are asymptomatic. Risk factors include exposure to ionizing radiation, inherited genetic syndromes, and certain environmental and lifestyle factors.
Diagnosis has advanced through innovations in imaging, cytology, molecular testing, and artificial intelligence. Ultrasound remains a key diagnostic tool, while fine-needle aspiration cytology and molecular testing for alterations such as BRAF, RET, and RAS can support diagnosis and treatment selection.
Treatment is personalized according to cancer type, stage, and molecular profile. Surgery remains the primary treatment, while radioactive iodine therapy is commonly used for differentiated thyroid cancers following surgery. Thyroid hormone suppression therapy can help reduce recurrence risk. For advanced or iodine-resistant disease, targeted therapies such as tyrosine kinase inhibitors and mutation-specific inhibitors are being used.
Emerging Thyroid Cancer Drug Profiles
AL2846: Chia Tai Tianqing Pharmaceutical Group Co., Ltd.
AL-2846 is an investigational small-molecule kinase inhibitor developed by Chia Tai Tianqing Pharmaceutical Group Co., Ltd. It targets receptor protein-tyrosine kinases, aiming to block cancer cell growth. The current trials are focused on its use in iodine-refractory differentiated thyroid cancer, both alone and in combination with other therapies, as well as neurofibromatosis type 1 and pancreatic cancer. Currently, the drug is being evaluated in Phase III clinical development for the treatment of Thyroid Cancer.
Vemurafenib: Genentech
Vemurafenib (Zelboraf) is an oral, small-molecule BRAF kinase inhibitor. The BRAF protein is a key component of the RAS-RAF pathway involved in normal cell growth and survival. Mutations that keep BRAF in an active state may result in excessive pathway signaling, leading to uncontrolled cell growth and survival. Vemurafenib is designed to block the BRAF mutation and may help kill cancer cells carrying the mutation or stop tumor growth. Currently, the drug is in Phase II clinical development for the treatment of Thyroid Cancer.
AIC100: AffyImmune Therapeutics
AIC100 is an affinity-tuned, third-generation CAR-T cell therapy engineered to target ICAM-1 on advanced thyroid cancers, including anaplastic and poorly differentiated thyroid cancers. ICAM-1, found on the surface of cancer cells, acts as a binding site for LFA-1 on AIC100 CAR-T cells, facilitating recognition and destruction of malignant cells.
AffyImmune Therapeutics has selected anaplastic and advanced differentiated thyroid cancers as the initial indications for AIC100. The therapy has received Orphan Drug, Fast Track, and Regenerative Medicine Advanced Therapy designations from the FDA. Currently, AIC100 is in Phase I clinical development for the treatment of Thyroid Cancer.
