Beta Thalassemia Epidemiology Segmentation Across the 7MM
The report provides detailed epidemiological estimates based on:
Total prevalent cases of Beta-thalassemia Minor
Diagnosed prevalence of Beta-thalassemia
Type-specific diagnosed prevalent cases of Beta Thalassemia
Total treated cases of Beta-thalassemia
Key epidemiological insights include:
Approximately 300 million individuals globally carry the thalassemia trait. More than 1 million people are estimated to have non-transfusion-dependent thalassemia, while over 100,000 patients are living with transfusion-dependent thalassemia. In the United States, at least 1,200 individuals are affected by transfusion-dependent thalassemia.
In the United Kingdom, the prevalence of beta-thalassemia major has been reported at approximately 0.3 per 100,000 population, while the National Organization for Rare Disorders (NORD) estimates the incidence of symptomatic cases in the United States at roughly 1 per 100,000 individuals.
According to the National Haemoglobinopathy Registry (NHR) 2020 Annual Report, the United Kingdom had more than 900 patients diagnosed with beta-thalassemia major and over 200 patients with beta-thalassemia intermedia.

Understanding Beta Thalassemia and Its Treatment Landscape
Beta-thalassemia is an inherited blood disorder resulting from mutations in the HBB gene on chromosome 11, leading to reduced or absent production of beta-globin chains in hemoglobin. The disorder causes ineffective red blood cell production, chronic hemolytic anemia, and impaired oxygen transport. Depending on disease severity, Beta-thalassemia is classified into minor, intermedia, and major forms.
The most severe form, Beta-thalassemia major (Cooley's anemia), is characterized by profound anemia, delayed growth, skeletal deformities, hepatosplenomegaly, and lifelong dependence on blood transfusions combined with iron chelation therapy. Additional disease variants include HbE/β-thalassemia, sickle β-thalassemia, dominant beta-thalassemia, hereditary persistence of fetal hemoglobin (HPFH), X-linked thrombocytopenia with thalassemia (XLTT), and TTD.
Diagnosis typically involves complete blood count (CBC), peripheral blood smear, hemoglobin electrophoresis or HPLC, molecular genetic testing for HBB mutations, iron overload assessment, and genetic counseling for affected families.
Marketed Therapies for Beta Thalassemia
Exagamglogene autotemcel (CASGEVY) - Vertex Pharmaceuticals: A CRISPR/Cas9 gene-edited cell therapy approved in the United States (2024) for transfusion-dependent beta-thalassemia patients aged 12 years and older.
Luspatercept (REBLOZYL) - Bristol Myers Squibb: A first-in-class erythroid maturation agent approved in 2019 for treating anemia in adults requiring regular red blood cell transfusions
Betibeglogene autotemcel (ZYNTEGLO) - Bluebird Bio: A gene therapy approved in the United States in 2022 for eligible patients with transfusion-dependent beta-thalassemia.
Mitapivat (PYRUKYND): An oral pyruvate kinase activator that enhances erythropoiesis and improves hemoglobin levels in transfusion-dependent and non-transfusion-dependent alpha- and beta-thalassemia.
Emerging Beta Thalassemia Therapies
Several promising pipeline therapies are advancing through clinical development, including:
Etavopivat (FT-4202) - Novo Nordisk: An oral pyruvate kinase activator currently in Phase III development with an anticipated U.S. launch around 2030.
SP-420 - Pharmacosmos: An oral desferrithiocin-class iron chelator in Phase II clinical evaluation following its acquisition from AbFero Pharmaceuticals.

Recent Developments in Beta Thalassemia
June 2026: Vertex Pharmaceuticals presented pivotal data at the European Hematology Association (EHA) Congress demonstrating consistent efficacy and safety of Exagamglogene autotemcel in children aged 5-11 years with severe sickle cell disease and transfusion-dependent beta-thalassemia.
May 2026: Agios Pharmaceuticals received European Commission marketing authorization for Mitapivat to treat anemia associated with both transfusion-dependent and non-transfusion-dependent alpha- and beta-thalassemia in adults, making it the first approved therapy in the European Union for this broad patient population.
2026: Novo Nordisk continued advancing Etavopivat, with Phase II clinical trial results anticipated during the second quarter of 2026.
Beta Thalassemia Market Outlook
Management strategies for Beta-thalassemia are primarily determined by disease severity and transfusion requirements. Regular blood transfusions remain the standard of care for patients with beta-thalassemia major and severe intermedia. Since repeated transfusions often result in iron overload, iron chelation therapies such as deferasirox, deferiprone, and desferrioxamine are widely utilized.
Supportive care also includes monitoring complications, managing splenomegaly, and performing splenectomy when clinically indicated. Hematopoietic stem cell transplantation remains the only established curative treatment for selected patients, although donor availability and transplant-related risks continue to limit its widespread use.
The therapeutic landscape is rapidly evolving with innovative therapies including CASGEVY, REBLOZYL, and ZYNTEGLO, while late-stage pipeline candidates such as Etavopivat, SP-420, and REGN7999 are expected to further transform the market by reducing transfusion dependence and addressing significant unmet clinical needs.
