What is Metastatic Castration-Resistant Prostate Cancer (mCRPC)?
Metastatic castration-resistant prostate cancer (mCRPC) is an advanced form of prostate cancer in which the disease continues to progress despite low testosterone levels achieved through androgen deprivation therapy. The disease spreads beyond the prostate gland to other parts of the body including bones, lymph nodes, liver, lungs, bladder, and in some cases, the brain.
Symptoms of mCRPC include trouble urinating, pain while passing urine, blood in urine, bone pain, fatigue, weakness, weight loss, and shortness of breath. Risk factors include advanced age, family history of prostate cancer, BRCA1/BRCA2 genetic mutations, elevated prostate-specific antigen (PSA) levels, and resistance to androgen deprivation therapy.
Diagnosis is primarily based on medical history, PSA testing, imaging procedures such as MRI and transrectal ultrasonography, and prostate biopsy. Treatment options include chemotherapy, immunotherapy, second-line hormone therapies, and radiopharmaceutical therapies.

PNT2002: Eli Lilly and Company
PNT2002 is a radioconjugate composed of a human prostate-specific membrane antigen (PSMA)-targeting ligand conjugated to the beta-emitting radioisotope lutetium Lu 177. The therapy targets and binds to PSMA-expressing tumor cells and destroys them through targeted beta particle radiation delivery. The therapy is currently being evaluated in Phase III clinical development for metastatic castration-resistant prostate cancer.

Onvansertib: Cardiff Oncology
Onvansertib is an oral polo-like kinase 1 (PLK1) inhibitor being developed for metastatic castration-resistant prostate cancer. The therapy is currently in Phase II clinical development and is being investigated for its ability to inhibit cancer cell proliferation and tumor progression.
