What is Metastatic Uveal Melanoma?
Metastatic uveal melanoma is an advanced form of uveal melanoma, a rare eye cancer originating from pigment-producing cells in the uveal tract, which includes the iris, ciliary body, and choroid. While primary uveal melanoma initially remains confined to the eye, nearly half of patients eventually develop metastatic disease, with the liver representing the most common site of metastasis, followed by the lungs and bones. Metastatic uveal melanoma is associated with poor prognosis due to limited effective treatment options and resistance to conventional chemotherapy and immunotherapy approaches. Current management strategies focus on systemic therapies, liver-directed interventions, and participation in clinical trials to improve patient outcomes and quality of life.

Emerging Metastatic Uveal Melanoma Drug Profiles

DYP688: Novartis
DYP688 is an investigational antibody-drug conjugate (ADC) administered intravenously that selectively targets metastatic uveal melanoma cells harboring activating GNAQ or GNA11 mutations. The therapy consists of an antibody directed against PMEL17 linked to a cytotoxic payload designed to inhibit oncogenic GNAQ/GNA11 signaling pathways responsible for tumor proliferation.
DYP688 is currently being evaluated in a Phase I/II clinical trial assessing safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with metastatic uveal melanoma and other GNAQ/11 mutant melanomas. Preliminary data demonstrated encouraging anti-tumor activity with favorable safety and tolerability profiles.

Darovasertib: IDEAYA Biosciences/Servier
Darovasertib is an investigational oral protein kinase C (PKC) inhibitor being developed by IDEAYA Biosciences in partnership with Servier for metastatic uveal melanoma. The therapy is currently in Phase II/III development and targets protein kinase C signaling pathways implicated in uveal melanoma progression.

Darovasertib is being evaluated both as monotherapy and in combination with crizotinib in registration-enabling studies for first-line metastatic uveal melanoma treatment.

Recent Developments in the Metastatic Uveal Melanoma Clinical Trial Landscape
• In December 2025, IDEAYA Biosciences announced completion of targeted enrollment of 435 patients in the registration-enabling Phase II/III OptimUM-02 trial evaluating darovasertib in combination with crizotinib for first-line HLA*A2-negative metastatic uveal melanoma. The company expects to report progression-free survival data in the first quarter of 2026 to support potential accelerated approval in the United States.
• In October 2025, IDEAYA Biosciences reported the first median overall survival results from the Phase I/II OptimUM-01 trial evaluating darovasertib in combination with crizotinib as a first-line treatment for metastatic uveal melanoma.
• In July 2022, Novartis initiated a Phase I/II multicenter study evaluating DYP688 in patients with metastatic uveal melanoma and other GNAQ/11 mutant melanomas.
• As of October 2024, preliminary clinical findings from DYP688 demonstrated confirmed objective responses, stable disease outcomes, and manageable adverse events in heavily pretreated metastatic uveal melanoma patients.
