What is Glioblastoma?
Glioblastoma multiforme (GBM) is the most aggressive and common form of malignant primary brain tumor in adults and is classified as a World Health Organization (WHO) grade IV astrocytoma. It originates from astrocytic glial cells and is characterized by rapid proliferation, diffuse infiltration, necrosis, and extensive microvascular proliferation within the central nervous system. Despite treatment advances involving surgery, radiotherapy, and temozolomide chemotherapy, the disease continues to have a poor prognosis, with median survival generally ranging between 12 and 15 months.

Symptoms of glioblastoma commonly include persistent headaches, nausea, vomiting, seizures, neurological deficits, cognitive dysfunction, memory impairment, and personality changes depending on tumor location and progression.

Diagnosis generally relies on MRI imaging, biopsy, histopathological examination, and molecular testing for biomarkers such as IDH mutation, MGMT methylation, and EGFR amplification.

Paxalisib: Kazia Therapeutics
Paxalisib is an orally administered phosphatidylinositol 3 kinase (PI3K) inhibitor being developed by Kazia Therapeutics for glioblastoma treatment. BPM31510: BPGbio
