In pathological conditions, this mid-region of tau plays a critical role in tau aggregation and is a key factor driving neurodegeneration in Alzheimer's disease. APOε4 is the primary genetic risk factor for sporadic Alzheimer's. At the 2025 CTAD conference, UCB presented promising data from the Phase 2a study of Bepranemab in early Alzheimer's disease. The results provided the first evidence of both biological and clinical effects of a mid-domain tau-targeting therapy aimed at modifying the disease. Although the primary endpoint was not met across the entire study population, Bepranemab demonstrated a slowing of cognitive decline and tau accumulation in key secondary endpoints. Consistent treatment benefits were observed in pre-defined patient subgroups across multiple primary and secondary measures, including cognition and function. UCB is now evaluating the next steps in the development of Bepranemab, which is currently in Phase II for Alzheimer's treatment.
