What is Open-Angle Glaucoma?

Open-angle glaucoma is a chronic, progressive, and irreversible optic neuropathy and represents the most common form of glaucoma. It is characterized by an anatomically open anterior chamber angle along with structural changes to the optic nerve head and retinal nerve fiber layer. In open-angle glaucoma, the drainage angle between the cornea and iris remains open, but impaired aqueous humor outflow can contribute to increased intraocular pressure and progressive optic nerve damage.
Open-angle glaucoma can be classified into primary and secondary forms. Primary open-angle glaucoma has no obvious underlying cause, with age-related changes in the trabecular meshwork contributing to increased resistance to fluid drainage. Secondary open-angle glaucoma occurs when another condition contributes to or accelerates dysfunction of the drainage system.
Symptoms may include gradual loss of peripheral vision, difficulty seeing objects at the edges of the visual field, missing letters while reading, and reduced visual contrast in low-light conditions. However, many individuals may not notice changes during the early stages, allowing the disease to progress silently and cause irreversible vision loss.
Treatment primarily aims to lower intraocular pressure and prevent further optic nerve damage. Management may include eye drops such as prostaglandin analogs, beta-blockers, and carbonic anhydrase inhibitors. When medical treatment is insufficient or poorly tolerated, laser trabeculoplasty and surgical interventions, including trabeculectomy, glaucoma drainage devices, and minimally invasive glaucoma surgeries, may be used.

HUC3-637: Huons Co., Ltd.
HUC3-637 is an investigational ophthalmic drug developed by Huons Co., Ltd. for the treatment of primary open-angle glaucoma and ocular hypertension. It is a fixed-dose combination of dorzolamide, a carbonic anhydrase inhibitor, and timolol, a beta-blocker, formulated as a topical eye drop. The drug is designed to reduce intraocular pressure by decreasing aqueous humor production through dual mechanisms. HUC3-637 is currently being evaluated in Phase III development for Open-Angle Glaucoma.

QLS-111: Qlaris Bio, Inc.
QLS-111 is a novel formulation based on an ATP-sensitive potassium (KATP) channel modulator platform. The drug is designed to reduce intraocular pressure by relaxing blood vessels and vascular-like tissues located distal to the trabecular meshwork. This mechanism is intended to decrease distal outflow resistance and lower episcleral venous pressure, which contributes to overall intraocular pressure regulation. QLS-111 has demonstrated sustained IOP reduction as monotherapy and in combination with standard treatments such as latanoprost. It is currently being evaluated in Phase II/III development for Open-Angle Glaucoma.

Xelafaslatide (ONL1204): ONL Therapeutics
Xelafaslatide (ONL1204) is a first-in-class small peptide that inhibits the Fas receptor. By blocking activation of the Fas pathway and related immune signaling, the therapy is designed to inhibit downstream cell-death signaling and inflammatory responses associated with retinal cell damage. This approach may help preserve retinal ganglion cells and other critical retinal cells. Xelafaslatide is currently being evaluated in Phase II development for Open-Angle Glaucoma.

IVW-2001 (GVB-2001): iVIEW Therapeutics, Inc.
IVW-2001 (GVB-2001) is an investigational gene therapy candidate developed by iVIEW Therapeutics for primary open-angle glaucoma. The therapy utilizes an optimized self-complementary AAV vector delivered through a single intracameral injection directly to the trabecular meshwork, which is a key anatomical site involved in aqueous outflow resistance. By modulating cellular function and enhancing aqueous humor outflow, GVB-2001 is designed to provide rapid and durable intraocular pressure control. It is currently being evaluated in Phase I/II development for Open-Angle Glaucoma.
