What is Hodgkin Lymphoma?
Hodgkin lymphoma is a cancer that starts in the lymphatic system, defined by the presence of Reed-Sternberg cells. It is one of two main types of lymphoma, the other being non-Hodgkin lymphoma, and is known for its high cure rates when detected and treated early. In Hodgkin lymphoma, B-lymphocytes multiply abnormally and accumulate in areas like the lymph nodes, losing their ability to fight infections, which increases vulnerability to illness. The two main types are classic Hodgkin lymphoma and nodular lymphocyte-predominant Hodgkin lymphoma. While Hodgkin lymphoma is aggressive and can spread quickly, it is also one of the most treatable cancers.

Hodgkin Lymphoma Approved Therapies
• KEYTRUDA (pembrolizumab): Merck
KEYTRUDA is a humanized monoclonal antibody and anti-PD-1 therapy that blocks the interaction between PD-1 and its ligands, PD-L1 and PD-L2, thereby activating T lymphocytes to help detect and fight tumor cells. In October 2020, the US FDA approved an expanded label for KEYTRUDA as monotherapy for adult patients with relapsed or refractory classical Hodgkin lymphoma, granted under accelerated approval based on data from the KEYNOTE-087 trial, which showed an overall response rate of 69%, including a 22% complete remission rate.

• OPDIVO (nivolumab): Bristol Myers Squibb
OPDIVO is a human IgG4 monoclonal antibody that binds to the PD-1 receptor, blocking its interaction with PD-L1 and PD-L2 and releasing PD-1 pathway-mediated inhibition of anti-tumor immune response. In May 2016, the US FDA granted accelerated approval to OPDIVO for classical Hodgkin lymphoma, having earlier received Breakthrough Therapy Designation and Orphan Drug status for relapsed or refractory disease.

• ADCETRIS (brentuximab vedotin): Pfizer & Takeda
ADCETRIS is an antibody-drug conjugate comprising an anti-CD30 monoclonal antibody linked to the microtubule-disrupting agent MMAE via a protease-cleavable linker, designed to release MMAE upon internalization into CD30-positive tumor cells. It holds marketing authorization in more than 70 countries for relapsed or refractory Hodgkin lymphoma. In November 2022, the FDA approved ADCETRIS in combination with a multidrug chemotherapy regimen for pediatric patients 2 years and older with previously untreated high-risk classical Hodgkin lymphoma, marking the first pediatric approval for brentuximab vedotin.

Hodgkin Lymphoma Pipeline Analysis
• TEVIMBRA (tislelizumab): BeiGene
TEVIMBRA is a monoclonal antibody targeting the PD-1/PD-L1 pathway, removing inhibition of the immune response. In August 2024, BeiGene reported completion of a Phase II trial (TIRHOL) in participants with relapsed or refractory classical Hodgkin lymphoma, and in March 2024 reported a separate Phase II trial evaluating tislelizumab as a first-line treatment in patients aged 60 and older, expected to complete in 2028.

• Favezelimab/pembrolizumab: Merck
This co-formulation combines favezelimab, which binds LAG3 on tumor-infiltrating lymphocytes to block its interaction with MHC class II, with pembrolizumab, and is being evaluated in a Phase III trial for relapsed or refractory classical Hodgkin lymphoma. Phase I/II results presented at ASCO 2024 showed an overall response rate of 83%, a 37% complete response rate, and a median progression-free survival of 19.4 months in anti-PD-1-naive patients.

Hodgkin Lymphoma Companies
Merck, Bristol Myers Squibb, Takeda, Seagen, Pfizer, ADC Therapeutics, Genmab, BeiGene, Tessa Therapeutics, AstraZeneca, and others.
