What is Chondrosarcoma?
Chondrosarcoma (CS) is a group of malignant bone tumors composed of cells that produce excessive cartilage. It is the second most common primary bone cancer and primarily affects the cartilage cells of the femur, pelvis, arm, knee, and occasionally the ribs.
The disease commonly presents with slowly progressive pain, swelling, restricted joint movement, stiffness, and in some cases neurological symptoms resulting from tumor compression. Chondrosarcoma is classified into multiple grades ranging from low-grade tumors with excellent prognosis to highly aggressive metastatic disease associated with poor clinical outcomes.
Diagnosis typically involves imaging techniques such as X-ray, CT, and MRI, followed by biopsy confirmation. Surgical excision remains the primary treatment approach, while chemotherapy and radiotherapy generally show limited effectiveness in most patients.

Emerging Chondrosarcoma Drug Profiles

Ivosidenib: Servier Bio-Innovation LLC
Ivosidenib is an oral small-molecule inhibitor that selectively targets mutant IDH1, a key molecular driver implicated in several cancers, including Chondrosarcoma. By inhibiting mutant IDH1 activity, the therapy reduces production of the oncometabolite 2-hydroxyglutarate and promotes normal cellular differentiation. Currently, Ivosidenib is being evaluated in Phase III clinical development for Chondrosarcoma.
The ongoing CHONQUER Phase III study is evaluating Ivosidenib in patients with locally advanced or metastatic IDH1-mutated conventional Chondrosarcoma and is expected to enroll approximately 136 participants.

Ozekibart (INBRX-109): Inhibrx Biosciences, Inc.
Ozekibart is a novel therapeutic candidate targeting human death receptor 5 (DR5) and is currently being evaluated in Phase II clinical development. The therapy represents an innovative targeted treatment strategy designed to induce tumor cell death and improve outcomes in patients with advanced Chondrosarcoma.

Recent Developments in the Chondrosarcoma Clinical Trial Landscape
• In January 2025, Servier announced enrollment of the first patient in the pivotal CHONQUER Phase III clinical trial evaluating TIBSOVO (Ivosidenib) versus placebo in patients with IDH1-mutated conventional Chondrosarcoma.
• In July 2024, Servier Bio-Innovation LLC initiated a Phase III multicenter, randomized, double-blind, placebo-controlled study evaluating Ivosidenib in patients with locally advanced or metastatic Chondrosarcoma.
• In October 2025, Inhibrx Biosciences announced positive topline results from the registrational ChonDRAgon study evaluating Ozekibart (INBRX-109) in advanced or metastatic unresectable Chondrosarcoma. The study enrolled 206 patients and demonstrated encouraging clinical outcomes.
• In October 2025, Transcenta Holding announced that, based on positive ChonDRAgon study results, Inhibrx intends to pursue regulatory filing for Ozekibart in the United States by the end of June 2026.
