What is Duchenne Muscular Dystrophy (DMD)?
Duchenne Muscular Dystrophy (DMD) is a severe genetic disorder characterized by progressive muscle degeneration and weakness. It primarily affects males and is caused by mutations in the DMD gene, which encodes for the protein dystrophin, essential for muscle cell structure and function. The absence or malfunction of dystrophin leads to the gradual breakdown of muscle tissues over time. It is the most common hereditary neuromuscular disease and does not exhibit a predilection for any race or ethnic group. The symptoms include muscle weakness, delayed motor development, enlarged calf muscles, toe walking, and waddling gait. The treatment involves corticosteroids, physical therapy, respiratory care, cardiac management, and exon-skipping therapies. The causes are associated with mutations in the dystrophin gene. The diagnosis is based on genetic testing, creatine kinase testing, muscle biopsy, and electromyography.

Emerging DMD Drug Profiles

CAP-1002: Capricor Therapeutics
Deramiocel (CAP-1002) consists of allogeneic cardiosphere-derived cells (CDCs), a population of stromal cells that have been shown in preclinical and clinical studies to exert potent immunomodulatory, antifibrotic and regenerative actions in dystrophinopathy and heart failure. CDCs act by secreting extracellular vesicles known as exosomes, which target macrophages and alter their expression profile so that they adopt a healing, rather than a pro-inflammatory, phenotype. The therapy is currently advancing through FDA review for Duchenne Muscular Dystrophy.

SGT-003: Solid Biosciences
SGT-003 is an investigational gene therapy being developed by Solid Biosciences for Duchenne Muscular Dystrophy (DMD). The therapy is currently in Phase III clinical development and is administered intravenously. The therapy is being evaluated for its role in treating Duchenne Muscular Dystrophy and improving dystrophin expression.
