Click Chemistry was first coined by Karl Barry Sharpless at the 217th ACS National Meeting in 1999.1 It refers to a chemical synthesis approach wherein molecular building blocks combine efficiently and selectively, generating no or fewer by-products that can be easily purified without chromatographic separation techniques. Notably, there are various types of click chemistry reactions; amongst these, bioorthogonal chemistry is most prominently employed in biological systems without altering the molecule of interest.Click and bioorthogonal reactions can be used in the production of therapeutic modalities and as probes in medical imaging techniques in order to aid in the development of treatment options for various oncological and non-oncological disorders. It is worth highlighting that in 2022, Carolyn R. Bertozzi, Karl Barry Sharpless and Morten Meldal were honored with "Noble Prize in Chemistry" for the development of click chemistry and bioorthogonal chemistry approach. A click reaction usually happens by combining an azide with a cyclooctyne, forming a conjugated biomolecule. The resulting molecule can be used as a probe to identify biomarkers in living beings.
