This old method required a separate slide for every single blood sample/concentration; analysing a small number of samples was a multi-week ordeal.
In a landmark innovation, Dr. Amadi optimised the Medium Throughput Alkaline Comet Assay, leading to his own branded comet assay method that has transformed the pace of research: The Amadi ProtocolTM or The Amadi Comet Assay ProtocolTM: An Optimised, Highly-Sensitive Medium Throughput Comet Assay for DNA Damage Assessment.
During his doctoral research at the University of Bradford, West Yorkshire, England, United Kingdom, in 2018, he was the first to introduce the Medium Throughput Comet Assay to the University of Bradford Toxicology Lab. of Professor Diana Anderson. Under the UK National Health Service (NHS) and around the world, Dr. Amadi was the first to investigate the genotoxic effects of commercially available graphene oxide (GO) nanomaterials (NMs) GO (15-20 sheets; 4-10% edge-oxidized; 1 mg/ml) in vitro, specifically, the DNA damage impacts and other genotoxic endpoints in human whole blood and peripheral blood leucocytes (PBL) from healthy individuals from Manchester and Bradford city, England and real-life patients diagnosed with chronic pulmonary diseases, i.e., asthma, chronic obstructive pulmonary disease (COPD), and lung cancer recruited from the ambulatory clinic of Professor Badie Jacob's Respiratory Clinic, Bradford Royal Infirmary (BRI) and Dr Abid Aziz, a Respiratory Consultant at St Luke's Hospital, Bradford, West Yorkshire, UK (2016-2019).
By the conclusion of his PhD in 2019, Dr Amadi had already refined and optimised the classical Comet Assay methodology in four core areas, resulting in a significantly improved technique. The classical decade-old Comet Assay, originally developed to detect DNA strand breaks at the single-cell level, suffers from several limitations:
1.High resource demands.
2.Experimental variability.
3.Unrealistic exposure durations.
4.Insensitivity to subtle DNA lesions.
5.Suboptimal electrophoresis conditions.
Through systematic refinement, Dr. Amadi addressed each of these weaknesses and transformed the assay into a more sensitive, ecologically and economically relevant method. The Amadi ProtocolTM is now suitable for translational studies in nanotoxicology and cancer research and has been recognised by independent experts, adopted by international laboratories, and is on track to become a new industry standard.
This new method allows for the simultaneous analysis of 12 samples on a single slide, compressing a two-week process into a single day. This extraordinary 93%-time savings is more than just a convenience; it allows for higher-volume experiments and significantly accelerates the pace of scientific discovery.

Dr. Amadi's preeminent standing in the field was recently cemented by the prestigious "Best Paper Award" he received in September 2024 from the International Journal of Health and Clinical Research (IJHCR) - Netherlands. This distinguished honour is granted to just one paper annually from a highly competitive pool of international submissions.
