CD BioSciences provides multi-color immunohistochemical staining kits that leverage either ABC (avidin-biotin complex) or polymer signal amplification techniques, facilitating the staining of tissues and cells in 2-3 distinct colors. These innovative kits are engineered to deliver superior quality and are specifically optimized for use with human and rodent samples. The new DS IHC Kits are compatible with a wide range of human and rodent tissues, including formalin-fixed, paraffin-embedded (FFPE) tissues and frozen tissues. For example, the 2-Color IHC Kit for 2 Rabbit Antibody on Human/Rodent Tissue (Catalog NO. MCIS063) is a versatile tool for immunohistochemistry, suitable for a variety of sample types including paraffin-embedded tissue, frozen samples and freshly prepared monolayer cell smears. Double staining is a common technique in immunohistochemistry that allows the simultaneous detection and differentiation of two different antigens within a single tissue sample. This kit is specifically designed for double staining applications and uses DAB (brown) and Fast Red (red) dyes to visualize two different antigens on human and mouse tissue or cell samples when combined with user-supplied rabbit primary antibodies.
In addition, the kit contains essential components for successful double staining. These include two polymer-enzyme conjugates: HRP Polymer Anti-Rabbit IgG and AP Polymer Anti-Rabbit IgG. These conjugates are coupled to two chromogenic substrates: DAB (used with HRP polymer anti-Rabbit IgG) and Fast Red (used with AP polymer anti-Rabbit IgG), which produce brown and red staining, respectively. The kit uses a non-biotin system to minimize non-specific binding.
The 2-Color IHC Kit for Goat/Rat Antibody on Human/Mouse Tissue (Catalog NO. MCIS070) uses two primary antibodies, goat and rat, to target two different antigens. To visualize these antigens, the kit contains two polymer-enzyme conjugates: HRP polymer anti goat IgG and AP polymer anti rat IgG. These conjugates are coupled to two different chromogens: AEC (red) and BCIP/NBT (purple). Sequential application of these conjugates allows the kit to detect both antigens simultaneously. If both antigens are present, a distinct purple/red colour develops. However, if only one antigen is present, either red or purple staining will be observed. Importantly, this non-biotin system minimizes background noise and ensures specific staining.
