One or two guide RNAs and Cas9 protein can be injected to generate knockout mice. Development of large knock-ins (approximately 5 to 10 kb long) is generally achieved using plasmid DNAs as donors. CRISPR editing or ESC-mediated gene targeting can be used to create humanized knock-in (KI) mice models; each technique has its advantages for model development.
