Head and neck cancer is a collective group of diseases comprised of laryngeal, oral and oropharyngeal cancers

Squamous cell carcinoma (SCCHN) accounts for more than 85% of all incidences of head and neck cancer. While tobacco and alcohol use have historically been associated with SCCHN, such cases have been on the decline. However, human papillomavirus (HPV) is now a well-accepted risk factor


First-line therapy in patients with the recurrent or metastatic disease is typically some combination of chemotherapy. Recommended treatment options include (1) cisplatin + 5-FU + cetuximab, (2) platinum chemotherapy + taxane or (3) cisplatin + 5-FU


Cetuximab, in combination with cisplatin and 5-FU, has established itself as the frontline standard of care for patients with recurrent or metastatic SCCHN. In addition to cetuximab, Merck’s pembrolizumab and Bristol’s nivolumab are the only other approved targeted therapies in SCCHN


AstraZeneca’s duralumin (anti-PD-L1 mAb) also has an extensive SCCHN program with multiple data readouts expected in 2018
