Optical Genome Mapping (OGM) is an advanced imaging technology that detects structural variations in DNA with high precision. It works by evaluating the fluorescent labeling pattern of individual DNA molecules, offering an unbiased, genome-wide assessment of structural variants as small as 500 base pairs (bp). This resolution far surpasses that of traditional cytogenetic techniques.

OGM employs a specialized DNA extraction protocol designed to isolate ultra-high molecular weight (UHMW) DNA. Unlike standard column-based methods, which can fragment DNA, OGM's paramagnetic disk system traps the DNA during washing steps, minimizing shearing forces. This process results in DNA fragments ranging from 150 kilobases (kb) to megabases (Mbp) in size, approximately 5-10 times longer than those obtained from conventional extraction methods. This larger fragment size allows for more comprehensive and accurate genomic analysis.
