Lennox Gastaut Syndrome Overview
According to National Organization for Rare Disorders (NORD; 2021), Lennox-Gastaut syndrome (LGS) is a severe form of epilepsy that typically becomes apparent during infancy or early childhood. Affected children experience several different types of seizures most commonly atonic, tonic, and atypical absence seizures. Children with Lennox-Gastaut syndrome may also develop cognitive dysfunction, delays in reaching developmental milestones, and behavioral problems. People with LGS begin having frequent seizures in early childhood, usually between ages 3 and 5. LGS is also a physically dangerous epilepsy syndrome of childhood because of the frequent falls, injuries, and cognitive impairment that can severely limit the quality of life. Children affected might previously have infantile spasms or underlying brain disorders, but the etiology can be idiopathic also. According to Epilepsy Foundation, Lennox Gastaut Syndrome accounts for only 2 to 5% of childhood epilepsies. In general, the causes of Lennox Gastaut Syndrome are divided into two broader groups i.e., Identifiable causes and Cryptogenic (non-identifiable) causes. Identifiable causes include brain damage (e.g., head trauma), perinatal complications (e.g., birth asphyxia, intrauterine growth retardation, kernicterus), congenital central nervous system malformations (e.g., tuberous sclerosis), infections (e.g., meningitis, sepsis), or metabolic disorders.

According to Lawrence et al., Epilepsy is relatively common, but its prevalence varies widely affecting approximately 3.4/1000 of the population in Japan, 6/1000 in the United States, and 11.2/1000 in Mexico.
According to Malik et al. LGS is a typical childhood-onset, severe epileptic encephalopathy associated with a serious intellectual disability in 20%-60% patients at the time of the onset of seizures, the proportion of which will increase to 75%-95% at five years after the onset of seizures.
According to NORD (2021), LGS affects males slightly more often than females. LGS is estimated to occur in 0.1-0.28 people per 100,000 and is believed to account for 1-4 percent of all cases of childhood epilepsy.

Lennox Gastaut Syndrome Treatment Market
Valproate is considered the first line of therapy for LGS as it is effective against a wide spectrum of seizures, the drug is administered as a monotherapy and if ineffective another drug such as lamotrigine, topiramate, rufinamide, or clobazam may be added. It also has significant side effects ranging from tremor to reduction in platelet count and function as well as hyperammonemia.
Several drugs have been approved by the US Food and Drug Administration (FDA) for the treatment of Lennox-Gastaut syndrome including Fenfluramine, Epidiolex, Topamax, Banzel, Lamictal, Felbatol, Onfi, and Klonopin. Interestingly, none of these are the typical first choice of treatment. The recently approved drugs are Fintepla (Fenfluramine) and Banzel (Rufinamide).
