Simply high dynamic binding capacities (DBC’s) are no longer sufficient. High DBC, at high flow rates with close to 100% recovery, across a wide range of molecular weights, this is what chromatographers look for in modern ion exchange resins. “BioPro” YMC’s hugely successful ion exchange material offers exactly this. The growing user base confirms very good separation, high dynamic binding capacities and excellent recoveries for the anion as well as the cation exchanger material.

The data below shows the DBC measured at high flow rates. The result: up to 1000 cm/hr the DBC stays very high. With measured amounts of at least 215 mg Lysozyme per ml of resin material, it reaches levels far above traditionally used cross-linked dextran and agarose gels. The graph below confirms that this high DBC is achievable across a wide range of flow rates. The graph shows the DBC of Lysozyme on YMC-BioPro S75 which is a sulphobutyl substituted cation exchanger with a particle size of 75 µm and a pore size of 100 nm (1000 Å). With its properties it is especially well suited for capture applications. However, at the same time, it is also usable for (intermediate) purification steps.

With a pore size of 100 nm the material is particularly useful for large molecules like monoclonal antibodies. However, tests with 5 µm analytical material show that YMC-BioPro can be used across a very wide range of molecular weights. The analytical material is based on the same hydrophilic resin material as the 75 µm preparative material and therefore shows very similar properties. The DBC for anion as well as cation exchanger material has been examined. The experiments cover a complete order of magnitude from roughly 14.000 to 150.000 Da. At the same time, this versatile material shows recoveries of generally 90% and more.

As a result, YMC-BioPro can help developing very economic bio-processes. With increasing titers of fermentations, high dynamic binding capacities are needed in order to keep column sizes at usable levels. High flow rates are needed to process the material in an acceptable amount of time. As shown above YMC-BioPro aids both points.

Additionally, with the knowledge gained from almost 30 years of experience with silica based chromatography phases, ÝMC succeeded to manufacture an IEX material with narrow particle and pore size distribution to warrant very good chromatographic performance.
